Experimental Drug TOFA Burns Fat Without Appetite Suppression
A Berkeley-led study found the experimental compound TOFA reduced obesity in mice by 18% in four weeks without altering food intake, physical activity, or body temperature—primarily burning fat while preserving lean mass.

An experimental compound has opened a new pathway for weight loss after demonstrating in obese mice that it increases fat burning without reducing food consumption, suppressing appetite, or causing muscle loss. The findings come from laboratory trials conducted by researchers at the University of California, Berkeley, and published in the journal Science Advances.
How TOFA Drives Weight Loss Without Behavioral Changes
Mice treated with TOFA—chemically named 5-tetradecyloxy-2-furoic acid—lost an average of 18% of their body weight over four weeks. Crucially, neither their food intake nor their physical activity levels changed during the treatment period. Body temperature also remained stable. Yet energy expenditure rose by up to 18%, indicating the compound prompted the body to burn more calories independently of dietary or behavioral adjustments.
The compound was administered twice daily to male obese mice for four weeks. Researchers observed that the majority of the weight lost consisted of fat mass, while lean body mass—including skeletal muscle and fat-free tissue—was largely preserved.
Mechanism of Action and Metabolic Improvements
TOFA appears to act through two simultaneous biological pathways: it reduces the synthesis of new fatty acids and enhances cellular uptake and oxidation of existing fats for energy production. However, the full mechanism underlying the observed increase in total energy consumption remains unclear.
Beyond weight reduction, TOFA-treated mice showed significant improvements across multiple metabolic markers: blood glucose and insulin levels declined, circulating lipids dropped, liver fat decreased, glucose tolerance improved, and several indicators of non-alcoholic fatty liver disease diminished.
Combination Therapy Outperforms Monotherapy
In separate experiments, low-dose TOFA was combined with two clinically approved weight-loss drugs—semaglutide and tirzepatide—both of which primarily function by suppressing appetite. The combination therapy resulted in greater weight loss and more pronounced improvements in metabolic parameters than either drug used alone, suggesting complementary mechanisms of action.
In another comparative trial, mice receiving TOFA maintained their reduced weight for a longer duration after treatment cessation. In contrast, mice treated with semaglutide rapidly regained weight, accompanied by increased food intake.
Human Trials Not Yet Conducted
The results remain preliminary. All experiments were carried out exclusively on a limited number of male obese mice. TOFA has not yet been tested in humans. Its efficacy, safety profile, and optimal dosing regimen in people are therefore unknown.
Researchers plan to conduct additional toxicology and efficacy studies in larger animal models before advancing to clinical trials in humans.
